In this case, it is a sense of belonging to a family routine that occurs daily at number 10 Mary Street. The mundanity of the routine provides stability and familiarity. Skrzynecki uses time frames such as â€Å"5pm† and â€Å"For nineteen years† to establish a sense of repetition and order in the reader’s mind. Collective pronouns such as â€Å"we† connote collaboration and inclusion in the family sphere. This family inclusivity allows the poet to establish his identity at an early age in a place in which he belongs, as shown when he describes him wandering in the garden after school. In the second stanza of the poem, Skrzynecki uses images of growth and nurturing to suggest a loving family environment and a sense of belonging to the land. The quiet â€Å"hum-drum† of daily routines, such as washing clothes and gardening, suggests that the house and Skrzynecki’s parents rarely change. This conjures an image of immense strength and solidarity. Skrzynecki establishes his childhood home as an enduring sphere of safety. He does this by personifying the house â€Å"in its china-blue coat† as a friend and part of the family. The home is a place in which to remember their Polish heritage. The repetition of the line â€Å"for nineteen years† illustrates the length of time that his family have been paying homage to their ancestry to as they â€Å"kept pre-war Europe alive. The use of the Polish word â€Å"Kielbasa† not only adds authenticity and depth to the poem but reinforces the idea that, though Skrzynecki’s family has moved away from war-torn Poland to Australia, they still firmly belong to their Polish heritage and there is a link for them and their family through which to establ ish their identities in their new land. The poet mourns the passing of his childhood and the destruction of the home in which he learnt the nature of growing up caught between two cultures and the rift between the past and the future. This notion is further explored in Apocalypse Now. Colonel Kurtz was the pride of the American Military Command. Having broken from the decrepit and corrupt school of thought that was the US army, Kurtz establishes his god-like rule over a clan of like-minded natives in the jungles of Cambodia. His character extrapolates all issues surrounding America as a nation, from war crimes to environmental stability. In one of the most compelling scenes of the film, Kurtz expresses his thoughts to Willard, one of the first Americans he has encountered since his dissent. He speaks of his son at home and his fear that if he were to be killed, his son would not understand his father’s actions. At this point, the extended close up shot of Kurtz’s face, half shrouded in darkness, changes slightly as he moves further into the light. This conveys that Kurtz still holds onto the hope that his son will one day come to understand his identity and why he acted in the way that he did. Kurtz is not ashamed of his actions because ultimately, he has fully formed his identity. First he was transformed on the battlefields of Vietnam by the death and ignorance he encountered/witnessed and then again in the jungles of Cambodia amongst the natives and free thought. Therefore, both 10 Mary Street and Apocalypse Now effectively explore the concept that acceptance and belonging are integral to the formation of one’s identity. A sense of belonging is achieved by following a path of alienation. In Migrant Hostel, Skrzynecki’s family struggle to establish themselves in a new land. Skrzynecki delineates the sense of alienation that the migrants have towards the rest of Australia. The â€Å"sealed off highway† demonstrates the separation they feel from the rest of the country. The simile of â€Å"rose and fell like a finger† demonstrates that they do not feel welcomed or accepted in their new land, but are constantly reprimanded, like a naughty child. The line â€Å"needing its sanction† demonstrates how the migrants are enslaved to the entrapment they feel in the hostel. They need permission to continue living in a manner that doesn’t reflect their culture or beliefs. This alienation from their culture and freedom renders each migrant unimportant and attempts to destroy their sense of personal identity and belonging. However, it is because of this alienation that they achieve a sense of belonging and identity. Nationalities ‘found each other’ based on their accents and the town they came from. Inside the hostel, they keep the memory of their home and culture alive though they are haunted by the â€Å"memories of hunger and hate† that destroyed their countries. Skrzynecki uses the simile â€Å"like a homing pigeon† to connote the strong sense of survival and solidarity shared by the migrants. The homing pigeon is a survivor that travels great distances. Skrzynecki uses a reoccurring motif of birds throughout this poem as they have connotations of freedom and migration. This dimension of belonging is further explored in John Steinbeck’s novel Tortilla Flat. The brothers eat, drink wine, sleep and occasionally venture out to do good deeds for those around them. They live by an entirely alternative concept of time, space, possession and love. The growing sense of belonging that develops through the novel is conveyed through the slow gathering of the six men to form the brotherhood and the corresponding rising action. Once they are all convened under a banner of bemused freedom, Danny states, â€Å"we are now as one, as never such men have been before. Each member is crucial to the group’s dynamic and therefore to each individual member’s sense of belonging. This is conveyed at the conclusion of the novel when, after Danny’s funeral, the house that was their home accidentally catches fire but instead of trying to save their one worldly possession, the men allow it to burn to the ground an d then go their separate ways. The last words of the novel are â€Å"no two walked together† conveying that the bonds of brotherhood had been broken and that it was only with each other that they belonged. Therefore, both Migrant Hostel and Tortilla Flat effectively convey the idea that belonging is reached by a path of alienation. Alienation leads to disillusionment with that which one once believed in (is there a different way to express this? ). Skrzynecki’s poem In The Folk Museum describes the experiences of the poet as he becomes increasingly alienated from his heritage. After describing his parent’s typical migrant experience in Migrant Hostel, the poet now finds himself unable to empathise with a past that is not his own. The use of first person not only allows the responder to connect on a deeper level with Skrzynecki, but also highlights the fact that he is alone in his musings about a past that he does not fully comprehend. In turn, this adds to the bleakness of an already melancholic poem. The caretaker of the museum represents everything that alienates Skrzynecki from his Polish heritage. She is knitting and has grey hair demonstrating that she is a relic herself and incongruent to contemporary society, just as Skrzynecki views his dying past. The simile of â€Å"cold as water† further illustrates that the poet no longer empathises or has any emotional connection to the events of his past. Although it is not as directly referred to in this poem(weak expression) as in others, In The Folk Museum also conveys how the poet’s disillusionment with his past leads to a sense of belonging with his present. The use of personification in â€Å"the wind taps hurriedly† communicates not only the poet’s frustration but also the determination of the outside world to remind him of the pointlessness of his reminiscing about his Polish heritage. The use of alliteration in â€Å"I leave without wanting a final look† conveys his speedy exit as well as his eagerness to regain the world outside of the museum, where he belongs. This notion of disillusionment is further explored in Francis Coppola’s film Apocalypse Now. Captain Willard, an American officer fighting in the Vietnam War, believes wholeheartedly in the US army and Western Civilisation as a whole. He belongs to war. At the beginning of the film, Willard is off duty in Saigon and recounts one of his trips home. The use of direct speech narration adds depth and authenticity to Willard’s character. He states â€Å"I’d wake up and there’d be nothing. Every minute I spend in this room, I get weaker and Charlie gets stronger. † The reference to the Vietcong warriors implies that Willard is more at ease when he is fighting in Vietnam. His alienation from normal society is further conveyed by a montage of images of war superimposed with Willard’s face. The non-digetic music of The End by The Doors plays, with lyrics such as â€Å"the west is the best† that further illustrates Willard’s faith in the American way of life and war. However, at the conclusion of this montage, Willard is left naked and bleeding, wrapped in a torn sheet and screaming on his hotel room floor, a high angle shot highlighting his vulnerability. This scene is purposely designed to alienate the audience from the character and connotes the detrimental effect that Western civilisation is having on him. The repetitious rigmarole of a soldier’s life is communicated through the undershot of the turning fan in Willard’s room. He stares up at it from the bed, implying that he is physically and mentally dominated by his life and routine as a soldier. As Willard travels further and further up river in search of Colonel Kurtz, he reads increasingly on Kurtz’s life and the events that have led him to the insanity that the US army now deems dangerous. Willard experiences more of the US army’s arrogance, blood-lust and drug use and becomes steadily disillusioned with the entity that he placed his faith in. Everything that is wrong with Western civilisation is represented through the arrogant Bill Kilgore who infamously states â€Å"I love the smell of Napalm in the morning. As the boat travels further up river, there is a distinct change in lighting. Before Willard boards the boat, there is a reoccurring motif of brightly coloured flares. The camera pans directly in front of the plumes of red, green and yellow smoke so that they form a veil over the scenes of battles and civilian deaths. However, once up river, the lighting becomes softer, greener and more defined. There is a distinct lack of smoke. This implies that Wi llard is travelling both physically and mentally away from the chaos of Western civilisation and heading deeper into Kurtz’s state of mind. Finally, Coppola uses the reoccurring motif of extreme close up shots on the faces of Willard and Kurtz. He does this to communicate that these two men are not necessarily similar but that they represent contrary facets of one human entity. The extreme close up shots of Willard and Kurtz reveal them to both be acutely serious men who have come to empathise with the same point of view. However, they are distinctly contrasted. Willard is often sweaty, dirty, constantly smoking whereas Kurtz is pristine, unchanging and aloof. This signifies that they will never be able to emulate the virtues of the other’s character that they themselves are deficient. Therefore, both In The Folk Museum and Apocalypse Now reveal themselves to be texts in which the view that alienation leads to disillusionment is explored. For humans to find where they truly belong, they must be placed outside of their comfort zone. They must travel beyond what they have before and thereby find something in the world, in others or in themselves that gives them a sense of belonging. She was not the driver of the car and the coffee was spilled while the car was parked. That is, Ms. Liebecks grandson had pulled the car to the curb so that Liebeck could add cream and sugar to her coffee and the vehicle was stationary before she placed the cup of coffee between her knees and attempted to remove the lid for the purpose of adding cream and sugar. As she lifted one side of the lid, the coffee spilled onto her lap. Immediately, the coffee was absorbed by her sweatpants. For home use, coffee is generally brewed at 135 to 140 degrees. If spilled on skin, any beverage heated to between 180 and 190 degrees will cause third-degree burns in two to seven seconds Ms. Liebecks injuries were severe. She suffered full thickness burns (third-degree burns) and scalding to her inner thighs, groin and buttocks. A vascular surgeon determined that Liebeck suffered full thickness burns over 6 percent of her body. She was in the hospital for eight days and had to undergo extremely painful procedures to remove layers of dead skin, as well as several skin grafting and debridement treatments (the surgical removal of tissue).Ms. Liebecks original intention was to obtain legal help in order to be reimbursed for her medical expenses, which were said to have totaled nearly $20,000. Tort law is easy to describe at a general level and hard to define more precisely. Tort comes from a Latin word meaning twisted or turned aside, so a tort is an act that is turned aside from the standard of proper conducta wrongful act. If you punch your neighbor in the nose, run over a pedestrian by driving carelessly, or injure a customer by serving burning hot coffee, you have committed a tort. (Some torts involve only economic harm and not physical injury, such as falsely accusing someone of being a crook or using fraud to induce them to enter into a financial transaction.) All of these are wrongful acts for which the victim can receive an award of money damages. Tort law permits private persons to take the lead in implementing its policies. All torts cases are brought by private individuals or businesses, not by the government acting as the prosecutor. What that means is fleshed out in the context of individual cases and can be hotly contested, but it does not require the law to specify in advance, in tedious detail, how a car must be built and what safety devices it must containThird, tort law links the deterrence and compensation policies to the objective of fairness by requiring that the compensation to the victim come from the wrongdoer. Once an inj ury has occurred, it seems right that the wrongdoer should be punished and the victim should be compensated. There is a neat symmetry to the mechanism that accomplishes both objectives at the same time. If the wrongdoer is criminally prosecuted, the victim still bears her loss; if the victim has her bills paid by insurance, her loss is compensated but the wrongdoer gets away without taking responsibility.During the trial process, McDonalds produced documents showing more than 700 claims by people burned by its coffee between 1982 and 1992. Some claims involved third-degree burns substantially similar to Liebecks. This history documented McDonalds knowledge about the extent and nature of this hazard. McDonalds also said during discovery that, based on a consultants advice, it held its coffee at between 180 and 190 degrees Fahrenheit to maintain optimum taste. McDonalds own quality assurance manager testified that a burn hazard exists with any food substance served at 140 degrees or a bove and that McDonalds coffee was not fit for consumption because it would burn the mouth and throat.The quality assurance manager further testified that the company actively enforces a requirement that coffee be held in the pot at 185 degrees, plus or minus five degrees. He also testified that while burns would occur, McDonalds had no intention of reducing the holding temperature of its coffee. Plaintiffs expert, a scholar in thermodynamics as applied to human skin burns, testified that liquids at 180 degrees will cause a full thickness burn to human skin in two to seven seconds. Other testimony showed that as the temperature decreases toward 155 degrees, the extent of the burn relative to that temperature decreases exponentially. Thus, if Liebecks spill had involved coffee at 155 degrees, the liquid would have cooled and given her time to avoid a serious burn. McDonalds also argued that consumers know coffee is hot and that its customers want it that way. The company admitted its customers were unaware that they could suffer third-degree burns from the coffee and that a statement on the side of the cup was not a warning but a reminder since the location of the writing would not warn customers of the hazard.A jury awarded Ms. Liebeck $200,000 in compensatory damages. This sum was reduced to $160,000 because, appropriately, she was found to be partially responsible for the incident. More significantly, the jury members saw fit to punish McDonalds. The jury awarded Ms. Liebeck $2.7 million in punitive damages. This amount represented approximately two days of profit for the restaurant chain. It also represented the single most publicized aspect of this case. The trial court subsequently reduced the punitive award to $480,000- or three times compensatory damages- even though the judge called McDonalds conduct reckless, callous and willful. Subsequent to remittitur, the parties entered a post-verdict settlement. Tort law is desiganted to protect consumer from unprincipled producer. In our case McDonald appeared to be careless as it many times before received claims r egarding the temperature of coffee. And as a result of McDonald’s negligence Ms. Liebeck burnt herself with too hot coffee. At the same time there is one point that makes this case bizarre. Everybody who buys coffee at McDonald’s knows that coffee is hot, very hot. This fact might have been the core of McDonald’s defense but it was neglected by the quantity of previous cases and awareness of the company management about such harmful facts.I must admit that the jury, after hearing all the facts and arguments taught McDonalds and other corporations a lesson: If you recklessly make or sell a dangerous product, you will be held accountable. McDonalds suffered substantial, but hardly outrageous, financial punishment for its irresponsible practices. Mrs. Liebeck was compensated for her injuries. The jury is always more eager to defend unprotected people such as old age, children, diasabled and so on.The jury’s decision helped not only Ms. Liebeck, after trial investigation discovered that McDonald’s reduced temperature of served coffee to 155 degrees; the case also influenced other potentially dangerous conduct: The Wendys chain reduced the temperature of its hot chocolate, served mostly to children. While critics of the tort system say that Liebeck vs MsDonald’s case was harmful to business, they claim that consumer shall be also responsible for the actions that lead to traumatizing. They say if, for example, a person burn himself preparing tea according to the instruction on the te a box, will he have reason to sue the Tea Company? It seems to be insane but it’d be the tea of the company that result burns. The only way to prevent such fabulous cases is developing of detailed instructions or warning for the customers to shift all preparation and consumption responsibility on them.If I were an owner of a small restaurant I would definitely change my serving policies. I would ask my waiters to warn customers that coffee, tea or other beverages are too hot, or I would decrease the temperature of serving beverages. How to Put Together a Poetry Manuscript for Publication Putting together a poetry manuscript to submit to contests or publishers is not a walk in the park. Expect it to take an hour or two a day over the span of a week, month, or even a year, depending on how much work you have, how polished the pieces are, and how much time you can afford to spend on the project.   Despite that, creating a poetry manuscript for publication is an important next step in a writers career. Heres a step-by-step guide on how to make this goal a reality. Step 1: Choose Your Poems Begin by typing (or printing from your computer files) all the poems you want to consider putting into your book, one per page (unless of course, the poem is longer than a single page). This is a chance to make any small revisions you want to make to individual poems so that you can concentrate on the shape of the book as a whole. Step 2: Plan the Book Size To get started, decide how big of a book you want to create- 20 to 30 pages for a typical chapbook, 50 or more for a full-length collection (more on exact page amounts later). You may well change your mind about this when you are actually selecting and ordering the poems, but this will give you a starting point. Step 3: Organize the Poems With the length of your book in mind, sift through all the pages you have typed or printed up, and put the poems into piles that you feel belong together in some way- a series of poems on related themes, a group of poems written using a particular form, or a chronological sequence of poems written in the voice of a single character. Step 4: Take a Step Back Let your piles sit at least overnight without thinking about them. Then pick up each pile and read through the poems, trying to see them as a reader and not as their author. You can use this time to mull over your reading, listening for the poems that stand out in each stack and how they sound together. Pay attention to other poems that may have popped into your mind when you were reading a certain stack to see if you should add them or replace similar poems. Step 7: Reevaluate Book Length Think again about the length of the book you want to create. You may decide that one stack of related poems would make a good short chapbook. You may have a really large pile of poems that will all go together into a long collection. Or you may want to combine several of your piles to create sections within a full-length book. Step 8: Create an Actual Book Next, try actually making the manuscript into a book that you can live with and leaf through. Staple or tape your pages together put them into a three-ring notebook, or use your computer to print them out in book format. Step 9: Choose a Title After you’ve decided on the length and general shape of your manuscript, choose a title for your collection. A title may have suggested itself during your sifting and ordering of the poems, or you may want to read through them again to find one- perhaps the title of a central poem, a phrase taken from one of the poems, or something completely different. Step 10: Proofread Carefully proofread your entire manuscript from beginning to end after you’ve put it in order. If you’ve spent a lot of time with the book, you may be tempted to give it only a cursory read-through. In this case, you need to set it aside for a few days or weeks so that when you come back to it you can pay close attention to each poem, each title, each line break, and each punctuation mark. You will likely find yourself making additional revisions to the poems at this point- don’t hold back, as this final reading may be your last chance to make changes before you send the book out into the world. Proofreading your own work is difficult- ask a friend, or two, to proofread the manuscript for you, and go through all their notes carefully. Fresh eyes will likely spot some errors that slid right by you but do not feel that you must accept every editorial change they may suggest. When in doubt about punctuation or line breaks, read the poem aloud. Step 11: Research Venues for Submission Next, it’s time to seek appropriate venues for submission. Use a list of poetry publishers or links to poetry contests to identify places you want to submit your manuscript. It’s important to read the poetry books they’ve published or the previous winners of their competitions in order to decide if you want them to publish your work. Targeting your submissions to publishers of like works can also save you time and money on submissions that would have been rejected for not being appropriate to their current catalog. Publishing is a business, and if a manuscript wouldnt fit in with others in the companys catalog, its marketing department wouldnt know what to do with it, regardless of its quality. Weed those publishers out before sending the manuscript anywhere. Keep notes on why a publisher is a good fit, to mention in your submission cover letter. Step 12: Apply! After you have selected a publisher or a contest, reread its guidelines and follow them exactly. Laboratory Report Essay The object of this experiment is to measure the effect of the concentration of the enzyme, amylase, on the rate of degradation of the starch and the effect of the chloride ion concentration in the solvent on enzyme activity. Also, the other object is to discuss the comparison between starch and cellulose digestion. Introduction: Starch is a polysaccharide and it is the major storage carbohydrate in plants where it is found in the plastids. It is made up of two types of polymers. One is amylose, the smaller linear and helical polymer, made up of many glucose molecules joined by ? 1 4 glycosidic bonds. The other larger polymer is amylopectin. It has a branched structure with many ? 1 4 glycosidic bonds between glucose molecules and ? 1 6 glycosidic bonds at branch points. Pure starch is in the form of a white powder which is tasteless and has no odor. It is insoluble in cold water. (Brown, W. H. Poon, T, 2005) Cellulose is another polysaccharide found in the cell walls of plants. It is made up of hundreds of linear chains of glucose molecules linked by ? 1 4 glycosidic bonds. Cellulose is a straight chain structure because the ? 1 4 linkage causes the glucose molecule to rotate 180 degrees. The many chains of cellulose are packed closely together by the hydrogen bonds existing between the hydroxyl groups of glucose molecules. These bonds contribute to the strength of the molecule. Cellulose is also odorless and tasteless and is insoluble in water (Young, Raymond ,1986). Starch is a major component of the human diet. For the digestion of starch, the enzyme amylase is needed which is present in the saliva and is also present in pancreatic secretions. This amylase breaks down starch into maltose, glucose, and limit dextrin. This enzyme works at an optimum pH of 6. 7-7. 0. Like for other enzymes, the greater the concentration of amylase present, the faster the digestion of starch would be. Also, chloride ions are coenzymes for this reaction and act as the allosteric activators of this enzyme. This means that the digestion of starch by amylase will only work in the presence of chloride ions (Thomas J. A. , Spradlin J. E. , Dygert S,1971). This experiment shows how the digestion of starch is carried out in the digestive tract by amylase and the conditions that are required for this reaction to take place. Method: to be inserted by the Results: a) This is a graph of the amount of the enzyme amylase in ml against the rate of reaction, taken as the reciprocal of the reaction time. Here, a linear relationship is seen and as the amount of the enzyme increases, so does the rate of the reaction. b) This is a graph of the chloride ion concentration in ? mol/tube against the rate of the reaction, taken as the reciprocal of the reaction time. This shows that as the concentration of chloride ions present increases, the rate of the reaction also increases. c) The ratio of the rates of digestion of these polysaccharides by salivary enzymes to bacterial enzymes can be approximately 10:1. This is because of the differences in the structures of starch and cellulose. Starch has just simple covalent bonds in its structure while cellulose has covalent as well as the stronger hydrogen bonds present to stabilize it. This is why it takes a longer time for bacterial enzymes to degrade cellulose compared to the time required to degrade starch. Discussion: The relationship that should exist between the rate of a reaction and the concentration of an enzyme should be a linear relation. This means that as the concentration of the enzyme increases, more active sites are present for the reaction to take place on, and so the overall reaction proceeds at a faster rate. The graph that we obtained from this experiment justifies this hypothesis because in it, there is a similar linear relation between the concentration of amylase and the rate of the reaction. After comparing the graph I obtained with two other students I realized that the enzyme that I used for my experiment was less active than the enzymes others used. It can be seen that when using the same amount of enzyme as the other students, the rate of my reaction is comparatively slower. For example, when using 5ml of enzyme solution of the same concentration, my reaction rate was around 0. 25 and others got a rate of around 0. 6 or more. That is greater than two times the difference in reaction rates. However, an error could have occurred if some of the enzymes supplied for the reaction were already denatured. In my graph of chloride concentration against the reaction rate, the shape of the curve is somewhat like this. In the beginning there is a great increase in the rate of reaction for a small increase in chloride concentration. However, as the concentration is further increased, the rate does not increase as much and the curve later flattens out. Humor in American Literature Essay

American society is unique, and the first of its kind. When Charles Dickens visited the United States he was astounded to see how informal American society was, as is recorded in his travelogue, American Notes For General Circulation. Besides this, it is often said by non-Americans that Americans have coarse senses of humor, or senses of humor that are low-class. The American sense of humor is said to clash with a polarized British one. The informality of American humor is utterly logical based on a theory that American society developed in response to Old-World British and European society. This theme often manifests itself in humorous American literature. Humor in American literature frequently bases itself on a sense of entitlement within the lower class, their flawed and ignorant moral senses, and a caricaturing of American exceptionalism, a theory of American superiority among other nations. American humor hasn’t changed extremely, but rather has maintained the common denominator of the humor of American exceptionalism and has parodied a feeling of individual self-worth that is seemingly intrinsic to Americans of all classes. This subject matter can be found in such works as Mark Twain’s The Adventures of Tom Sawyer, Sut Lovingood: Yarns Spun by a â€Å"Nat’ral Born Durn’d Fool†, by George Washington Harris, and Stephen Colbert’s I am America (and So Can You). Most of George Washington Harris’s work was done in the middle of the nineteenth century. His most famous character is the stereotypical rural farmer, Sut Lovingood. Harris’s Sut Lovingood stories were told from the character’s perspective and were defined by Sut’s heavy â€Å"Appalachian English† accent. His stories were initially published separately in multiple newspapers until Harris compiled the stories in his 1867 book Sut Lovingood: Yarns Spun by a â€Å"Nat’ral Born Durn’d Fool. The book mostly pokes fun at the ignorance and simplicity of the title character. Sut Lovingood dictates the book to a writer. He is unable to write it himself because he is illiterate. He is uneasy about this though. Sut says: â€Å"I don’t like the idear ove yu writin a perduckshun, an’ me a-findin the brains.† (Harris 2) Sut is an immensely simple character, but does not see himself as such. He thinks he has important things to say and that anyone who doesn’t read his book will simply be worse-off for having done so, but in reality his stories mostly consist of folderol. Sut states that he wanted his dedication page to read as such: â€Å"Dedercated wif the symperhtys ove the orthur, to the man ur ‘oman, huever they be, what don’t read this yere book.† (Harris 5) Despite his coming after George Washington Harris, Mark Twain is often hailed as the father of American Humor. Twain’s tales of life on the Mississippi River became the definitive stories of his era. His stories are often about naivetà © and the innocence of childhood, on a superficial level , but on a deeper level they are about ignorance and other shortcomings of adults. Twain’s stories were simultaneously entertaining and critical of society. One of his most famous works of this classification is The Adventures of Tom Sawyer. Tom Sawyer is set in a small Missouri town, where the standards for luxury are quite low, and the people easily impressed. The townspeople are awed by the county-Judge Thatcher, a relatively low figure in the governmental hierarchy. When he pays a visit to the town church everyone there bends over backwards to â€Å"show off† and impress him (Twain 38). The people are especially impressed by the fact that there exists a tin roof atop one of the buildings in Judge Thatcher’s town. Tom Sawyer’s Aunt Polly is an excellent caricature of American humor. Despite being the voice of discipline in Tom’s life, Aunt Polly is constantly being fooled by the boy. Among the most well known of the Tom Sawyer stories is the anecdote about doing the chore of whitewashing a fence. Because of his indolent nature, Tom cozens his peers into completing his task for him, unbeknownst to his aunt. After Tom returns from his supposed labor after only a short time, Polly commends him saying, â€Å"Well, I never! There’s no getting round it, you can work when you’re a mind to, Tom†. And she goes on to give Tom a â€Å"lecture upon the added value and flavor a treat took to itself when it came without sin through virtuous effort. And while she closed with a scriptural flourish, he ‘hooked’ a doughnut.† (Twain 26) Not only is Tom able to totally fool his aunt, but he is confident enough in her incompetence that he even snatches an additional snack. She and all of the adults in the story go on to fall for Tom’s wiles on numerous occasions. The more modern Stephen Colbert wrote a book entitled I Am America (and So Can You), which bears many similarities to George Washington Harris’s book. Colbert’s work is written from the character on his satirical news show’s extremely traditional, right wing perspective. He spoofs the idea of a traditional American family, going so far as to say that anything falling short of his expectations is simply wrong. He remarks on the role of a good, traditional mother: â€Å"A good mother cooks, cleans, drives, organizes charity events so her children earn community service points for college, and expects nothing in return except love and breakfast in bed one day a year.† (Colbert 9), and he lambasts the idea of one-parent families saying, â€Å"â€Å"If you ask me, from time immemorial, the word ‘family’ has meant only one thing: a Mom married to a Pop and raising 2.3 rambunctious little scamps.† (Colbert 5) Colbert is joking of course, but his implications are that the traditional American standards have room for adjustment and that many conservative Americans are overly hidebound. He goes on to lampoon the institution of religion. â€Å"Religion is the cornerstone of civilization. Without it, we would have no laws, no morality, no social structure, and no guidelines for furnishing our tabernacles.† Many people have tried to pinpoint exactly what it is so distinctive between American and British humor. British comedian Simon Pegg wrote about in the UK Guardian in his 2007 article, â€Å"What Are You Laughing At?†: Americans can fully appreciate irony. They just don’t feel entirely comfortable using it on each other, in case it causes damage. A bit like how we feel about guns. It’s not so much about having a different sense of humour as a different approach to life. More demonstrative than we are, Americans are not embarrassed by their emotions. They clap louder, cheer harder and empathise more unconditionally. It’s an openness that always leaves me feeling slightly guilty and apologetic when American personalities appear on British chat shows and find their jokes and stories met with titters, not guffaws, or their achievements met with silent appreciation, rather than claps and yelps. We don’t like them any less, we just aren’t inclined to give that much of ourselves away. Meanwhile, as a Brit on an American chat show, it’s difficult to endure prolonged whooping without intense, red-faced smirking. It isn’t that one culture is funnier per se, just that comparing the two would be like comparing apples and oranges. In former Vanderbilt English professor Nancy A. Walker’s book, What’s So Funny? Humor in American Culture, Walker expounds upon the origin of the distinctive American â€Å"tall tale†, citing the American pioneers of the Western frontier who would write back to the East Coast telling tales of exaggeration in an attempt to impress the readers of their tales. Edward J. Piacentino wrote that the purview of classic American humor still reaches us through mediums such as The Beverly Hillbillies, or the work of comedian Jeff Foxworthy. America’s unique face of humor stems from multiple factors of American history. Firstly, American humor developed intentionally and in stark contrast to that of the British, just like much of American society. Because of the revolutionary nature of the founding of America, certain values have been instilled in the country’s inhabitants. American culture developed as a volte-face from British culture. Additionally, as Walker presented, the development of American humor and exaggerated â€Å"tall-tales† derives from the geography of the country and the frontier development of it. Those elements of the country’s history laid the groundwork for the America that we know today. That is why we still have much of the same face of humor in America after all these years. Learn More About Inventer Thomas Elkins Dr. Thomas Elkins, an African-American inventor, was a pharmacist and respected member of the Albany community. An abolitionist, Elkins was the secretary of the Vigilance Committee. As the 1830s drew to a close and the decade of the 1840s began, committees of citizens were formed all across the north with the intention of protecting fugitive slaves from re-enslavement. As slave catchers sought fugitives vigilance committees provided legal assistance, food, clothing, money, sometimes employment, temporary shelter and assisted fugitives in making their way toward freedom. Albany had a vigilance committee in the early 1840s and into the 1850s. Thomas Elkins - Patents and Inventions An improved  refrigerator  design was patented by Elkins on November 4, 1879. He designed the device to help people have a way of preserving perishable foods. At that time, the common way of keeping food cold was to place items in a large container and surround them with large blocks of ice. Unfortunately, the ice generally melted very quickly and the food soon perished. One unusual fact about Elkins refrigerator was that it was also designed to chill human corpses. An improved chamber commode (toilet) was patented by Elkins on January 9, 1872. Elkins commode was a combination bureau, mirror, book-rack, washstand, table, easy chair, and chamber stool. It was a very unusual piece of furniture. On February 22, 1870, Elkins invented a combined dining, ironing table, and quilting frame. The Refrigerator Elkins patent was for an insulated cabinet into which ice is placed to cool the interior. As such, it was a refrigerator only in the old sense of the term, which included non-mechanical coolers. Elkins acknowledged in his patent that, I am aware that chilling substances enclosed within a porous box or jar  by wetting its outer surface is an old and well-known process.   Unique Folding Table A patent was also issued to Elkins on February 22, 1870, for a Dining, Ironing Table and Quilting Frame Combined (No. 100,020). The table seems to be little more than a folding table. The Commode The Minoans of Crete are said to have invented a flush toilet thousands of years ago; however, there is probably no direct ancestral relationship between it and the modern one that evolved primarily in England starting in the late 16th century, when Sir John Harrington devised a flushing device for his godmother Queen Elizabeth. In 1775, Alexander Cummings patented a toilet in which some water remained after each flush, thereby suppressing odors from below. The water closet continued to evolve, and in 1885, Thomas Twyford provided us with a single-piece ceramic toilet similar to the one we know today. In 1872, a U.S. patent was issued to Elkins for a new article of chamber furniture which he designated a Chamber Commode (Patent No. 122,518). It provided a combination of a bureau, mirror, book-rack, washstand, table, easy chair, and earth-closet or chamber-stool, which might otherwise be constructed as several separate articles.

Compare and Contrast the Development of B and T Cells The epithelial surfaces of the body serve as an effective barrier against most microorganisms, and they are rapidly repaired if wounded. Adaptive immunity is initiated when an innate immune response fails to eliminate a new infection, whereby an activated antigen presenting cells (APCs) bearing pathogens antigens are delivered to the draining lymphoid tissues. An adaptive immune response differs from the innate immunity in its ability to target structures that are specific to particular strains and variants of pathogen. T cells are produced in the bone marrow. They are transported still, as pro-thymocytes to the thymus where they undergo the process of maturation and selection. The regulation of T cell maturation in the thymus is termed central tolerance. During gestation, most T cells generated bear the gamma/deta T cell receptor (TcR) on their surface. In the adult, most T cells bear the alpha/beta TcR. The newly formed TcR then, has to be tested for recognition of self-MHC/peptide. The T cells are tested at a stage of development known as double positive, meaning that they bear both CD4 and CD8 receptors on their surface. Cells with TcRs that recognize self-MHC/peptide with very low affinity will die. This process is known as death by neglect. Cells with TcRs with medium affinity for MHC receive survival signals and undergo a process known as positive selection. Finally, cells which receive a high affinity signal via their TcR die by apoptosis, a process known as negative selection. Cells that in teract with MHC class I become CD8 positive T cell, and those that interact with MHC class II become CD4 positive T cells, before migrating out into the peripheral lymphoid system (Wood P, 2006). Mature B cells, like T cell, are also develop form pluripotent stem cells. However unlike T cells lymphocytes, B cell maturation occurs in the bone marrow. There are four different stages of B cell development: pro-B, pre-B, immature B, and mature B cells. During its development, B cells acquire B cell surface marker expression such as B220, CD19, CD20, etc. as well as antigen receptors. The stromal cells lining the bone marrow provide essential growth signals to developing B cells, including cytokines such as IL7 and cell to cell contact, via VLA4/VCAM and Kit/SCF. During B cell development, gene segment rearrangements take place, just like in T cells where TcR rearrangements (central tolerance) also occur. However, for B cells, the immunoglobulin heavy chain gene locus (variable-V, joining-J and diversity-D segments), situated on chromosome 14, rearranges. In haematopoietic stem cells, the Ig heavy chain genes are in germline configuration (Kurosaki T et al., 2009). As B cells deve lop to pro-B cells, a D-J recombination is the first gene rearrangement to take place. The intervening DNA is normally deleted from the chromosome as a circle. Gene rearrangements are mediated by recombinase activitng genes, RAG proteins. As the developing B cell proceeds from pro- to pre-B cell stage, a V-DJ gene arrangement takes place to form the VDJ coding block that encodes the variable domain on the antibody heavy chain. Gene rearrangement takes place on both copies of chromosome 14 in a developing B cell, but once a productive VDJ block has been assembled on one chromosome 14, rearrangement ceases on the other chromosome, ensuring only one type of Ig is produced by any single B cell. This process is known as allelic exclusion. If a developing B cell fails to make a productive VDJ block, it will fail to produce antibody heavy chain and die in the bone marrow (Murphy K et al, 2008). T and B cell activation: T cell activation takes place in draining lymph nodes (also spleen) close to site of infection. T cell recognizes antigen on MHC (Major Histocompatibility Complex) molecules becomes activated and differentiates to effector cells. Effector T cells migrate to site of infection and carry out effector functions. The T lymphocytes arrive through venules, and cross through the endothelial to the lymph nodes. Antigen presenting cells such (APC) such as dendritic cells, and macrophages presented antigens to T cells. On recognition of the antigen, a low affinity interaction is formed. These T cells then leave lymph node though the lymphatic system. Those T cells that recognize the antigens wall with high affinity will be retained and the process of proliferation and differentiation occurs. However, initial B cell activation takes place in T cell zone of secondary lymphatic tissues (i.e. in lyhmph nodes). Mostly IgM producing plasma cells are produced at this state. B cells, unlike T cells, ar e activated by the ineraction with antigen-specific T cell, by linked recognition. Antigen-activated B cell migrates to B cell area of lymph nodes to form organized germinal centres, where additional B cell differentiation processes take place. It is important to note that T cells recognize the peptide, while B cells recognize the coat protein. For T and B lymphocyte activation 2 signals are hypothesized to be required. Firstly, the antigen stimulus signal and secondly, the co-stimulatory stimulus. The absence of the second signal results in anergy or apoptosis. CD28/B7 interaction is the co-stimulatory signals for T cells while CD40/CD40 ligand, on the activated T cells, interaction is for B cells. For both T and B lymphocytes, in it resting G0 cell cycle, the cell appear to have a large nucleus, with little cytoplasm and show little evidence of organelles. However, when these cells enter G1/S/G2 cell cycle, cell shows an increase in cell size, chromatin de-condensation is seen. Cell division occurs rapidly, generating effector cells of either T or B lymphocytes. Effector T cells include Th1, Th2 and T regulatory, as well as T cytotoxic cell and memory T cells. On the other hand, effector B cells include plasma cell and memory B cell. T and B cell effector functions: B cell response to T-dependent protein antigen results in germinal centres formation in B cell areas of lymph nodes, and specialized processes such as Ig class-switching, somatic mutation and affinity maturation, memory B cell and plasma cell generation take place there. Emerging form germinal centres are somatically mutated and class-switched B cells, which no longer just produce IgM. Memory B cells are long-lived, resting and re-circulating cells, responsible for immunization part which helpto generate rapid and vigorous immune response on second encounter for that specific antigen. Plamablast cells migrate to other sites such as bone marrow, and become plasma cells, producing large amounts of secreted antibody. Some of which can live for long periods. The effector functions of B cells refer to what antibodies do after their contact with the antigen. The antibody effector functions include neutralization, complement fixation (IgM, IgG1/2/3), oposonization and antibody dependent cel l-mediated cytotoxicity. In contrast, T cell effector functions differ significantly from B cell effector functions. Antigen presenting cells present peptide via MHC which can either interact with CD4+ or CD8+ T cells. Helper T cells are defined by the cytokines they produce. Naà ¯ve CD4+ T cells (Th0), on interaction with APC, can differentiate to Th1 or Th2 cells, depending on the cytokine environment. Th1 cells co-ordinate inflammatory immune responses to intracellular pathogens while Th2 cells help B cells to make antibodies required for immune responses to extracellular pathogens, this is known as humoral immunity. Th1 and Th2 cells both act to promote the generation of more leukocytes. Besides Th0/Th1/Th2, other CD4+ T cell subsets exist (Zhu J et al., 2010). Resting T cells can differentiate into activated helper T cell, as well as activated cytotoxic T cell (CD8+ T cell). Initially, CD8+ T cells interact with potential target cells via low affinity/non-specific interactions between adhesion molecule s on the T cell (LFA-1 and CD2) and the target cell (ICAM1, ICAM2). This interaction has no effect on the cytoskeleton of the T cell and is a transient interaction unless recognition of specific peptide:MHC complexes occurs. If peptide:MHC I complex is present, the affinity of the adhesion molecule interaction increases and there is clustering of T cell receptor and associated molecules at the point of contact with the target cell forming the immunological synapse. This also signals for cytoskeletal rearrangements organized by the microtubule organizing complex which focuses the cytotoxic granules of the T cell at the point of contact with the target. Notice here, that T cells, unlike B cells do not produce antibodies against antigens. Granules containing perforin and other enzymes including granzymes are released and induce the activation of the cathepsin pathways in the target cell leading to apoptosis. CD8+ T cells can also kill target cells via the Fas/FasL pathway which also in duces apoptosis (Peter EJ 2007). In conclusion, adaptive immune responses occur when individual lymphocytes capable of responding to antigen proliferate and differentiate to become an antigen-specific effector cells and memory cells. The process of lymphocyte cell cycle progression, proliferation and differentiation in response to antigen and stimuli is known as lymphocyte activation. B cell activation is initiated by the ligation of the B cell receptor (BCR) with antigen and ultimately results in the production of protective antibodies against potentially pathogenic invaders. While naive or memory T cells encounter foreign antigen along with proper co-stimulation they undergo rapid and extensive clonal expansion. In human, this type of proliferation is fairly unique to cells of the adaptive immune system and requires a considerable expenditure of energy and cellular resources.
